Synthesis notes of Mescaline from 3,4,5-TMBA I update real time as I make a progress

Here I'll document my Mescaline synthesis from 3,4,5-trimethoxybenzaldehyde then after I optimize the reaction I'll write the documents.
My goal is to achieve good yield using minimum catalyst and nitromethane.

3,4,5-trimethoxybenzaldehyde in a bag

Experiment 1:​

10g of 3,4,5-trimethoxybenzalnyde was added to the 65ml of IPA which was stirred and heated to dissolve, 3.733g (1.2 mol eq.) of Nitromethane was added to the mixture, 0.374g (0.12 mol eq.) of Ethanolamine and 7.652g (2.5 mol eq. to Ethanolamine) of Acetic acid was mixed together then heated on a 130°C hotplate to make 2-HEAA then it was added to the mixture, The mixture was stirred and heated to 35°C for 3h then cooled to room temperature after about a hour there was needle looking yellow transparent crystal.

lol i have been trying to finesse this over the past year as well but there is only so much mescaline one man needs! Haha

I literally have a Henry reaction with nitroethane going as we speak. I am intending to resolve the enantiomers and comparatively bioassay them.

I never really think to write up my syntheses on here, I just presume meh, they’ve already been done…so I’m working on some that havn’t been done now (not so much the enantiomers, that has been done)

I also recently gassed out a load of acetone with HCl for the purpose of being able to salt out amine’s easier without having to gas or use aqueous HCl - I would recommend it then just keep a stock solution.

I did actually start by trying mescaline sulfate but my god do you need to swallow so much of the stuff. Not worth it
