Benzphetamine is metabolized by the human body
into amphetamine and methamphetamine,
[ prodrug of dextromethamphetamine and dextroamphetamine ]

Amphetamine and related sympathomimetic medications (such as benzphetamine) are thought to stimulate the release of norepinephrine and/or dopamine from storage sites in nerve terminals of the lateral hypothalamic feeding center, thereby producing a decrease in appetite. This release is mediated through the binding of benzphetamine to VMAT2 and inhibiting its function, causing a release of these neurotransmitters into the synaptic cleft through their reuptake transporters. Tachyphylaxis and tolerance have been demonstrated with all drugs of this class.

Materials Needed:
- Methamphetamine Hydrochloride: 10 g
- Potassium Carbonate: 11.60 g
- Benzyl Chloride: 4.61 g
- Water: 50 mL (reaction medium)
- Sodium Hydroxide Solution: (for pH adjustment to ~13)
- Naphtha: 50 mL per extraction (3 extractions total)
- Distilled Water
additional (for titration): 80 mL - 15% Hydrochloric Acid Solution: (sufficient quantity to adjust pH to 4-5)
- Isopropanol:100 mL
- Acetone: (sufficient for washing and crystallization)
Equipment Needed:
- Reaction vessel
- Magnetic stirrer with stir bar
- pH meter or pH indicator strips
- Separatory funnel
- Evaporating dish (Pyrex preferred)
- Filtration setup
Step-by-Step Procedure:
1. Dissolution of Starting Material:
- Dissolve 10 g of methamphetamine hydrochloride in 50mL of water in a clean reaction vessel at room temperature.
2. Addition of Potassium Carbonate:
- Slowly add 11.60 g of potassium carbonate to the solution while stirring.
- Continue stirring for 30 minutes to ensure complete reaction.
3. Benzylation Reaction:
- Add 4.61 g of benzyl chloride to the mixture.
- Heat the reaction mixture to 70-75°C and maintain this temperature for 10 hours with continuous stirring.
- let it cool to room temperature with additional stirring.
- Dilute with 100 ml cool water. stir and let it rest 5 minutes
4. Basification:
- After completion of the reaction, cooing and diluting the mixture,
- Adjust the pH to approximately 13 using sodium hydroxide solution, monitoring with a pH meter./strips
5. Extraction:
- Perform three consecutive extractions using 50 mL of naphtha for each extraction.
- Combine the naphtha layers in a separate vessel containing 80 mL of distilled water.
-> pour the Naphtha extracts on top of aqueous layer, (will stay on top...)
6. Acidification to Form Hydrochloride Salt:
- Equip the vessel with a magnetic stir bar and gradually titrate with 15% hydrochloric acid.
- Add the acid in small portions, stirring vigorously after each addition.
- Monitor the pH after each addition until it reaches 4-5.
- After reaching the desired pH, continue stirring vigorously for an additional 30 minutes to
ensure complete conversion of the freebase ( from Naphtha Layer) to the hydrochloride salt (then dissolved in the DH2O Layer!).
7. Isolation of the Hydrochloride Salt:
- Separate the aqueous layer containing the hydrochloride salt. (discard the Naphtha layer, store, to reuse , its perfectly fine again)
- Add 100 mL of isopropanol to the aqueous layer and concentrate the mixture by evaporation (to speed up evaping process!)
under ventilation with constant slight magnetic stirring in a Pyrex evaporating dish until most of the liquid has evaporated.
8. Crystallization:
- Remove the stir bar and perform flash crystallization by adding a sufficient amount of acetone.
- Collect the crude crystalline mass, dry it, and rinse three times with generous portions of acetone.
9. Recrystallization:
- Dissolve the washed salt conglomerate in a heated saturated isopropanol solution.
- Remove the solution from the heat source and allow it to stand undisturbed to promote slow crystallization.
10. Final Drying:
- Once crystallization is complete, rinse with cold acetone if necessary, and dry thoroughly
