Amphetamine Amphetamine synthesis by P2NP with NaBH4/CuCl2 ( Quantity 1 POT/1 KG )

Reaction scheme

Reagents


  • Isopropyl alcohol (IPA) - 20 L.
  • Distilled water - 6.2 L.
  • Sodium tetrahydruroborate (NaBH4) - 1739 g.
  • P2NP - 1000 g.
  • Copper(II) chloride - 105 g.
  • Sodium hydroxide (NaOH) 25% aqueous solution - 8 L.
  • Dry acetone - 2.5 L.
  • Sulphuric acid (or Phosphoric acid).

Equipment and Glassware


  • Batch reactor (measurement range: 50 L) equipped with reflux condenser, top stirrer and heating system;
  • Nutsche filter;
  • Laboratory balance (1-2000 g);
  • pH indicator paper;
  • Several buckets for 10 and 20 L;
  • Laboratory thermometer (measurement range: 150 °С);
  • Freezer;
  • Pyrex dishes for crystallisation;
  • Graduated test tube for 1 L;
  • Conventional funnel;
  • Drop funnel;
  • Vacuum pump;
  • 2 Beakers for 5 L;
  • Acid cold water bath for reflux condenser;

Synthesis procedures


  1. A 2:1 mixture of IPA and water (18 L) is added to the batch reactor.
  2. Solid NaBH4 (1739 g) is added to a portion and the stirrer is started.
  3. Pure P2NP (1000 g) is added in several small portions to keep the mixture below 60°С.
  4. A CuCl2 solution (105 g) in water (200 ml) is slowly added drop by drop to keep the mixture below 80°С.
  5. The reaction is then kept at 80 °C for 30 minutes with the help of an external radiator.
  6. NaOH (8 liters 25%) in aqueous solution is added to the chemical reaction and the layers are separated.
  7. The aqueous phase is extracted with 8 litres of IPA. The amphetamine is dissolved in the IPA and the density of the IPA layer changes as it is separated from the aqueous solution layer.
  8. The IPA extract from step 7 is evaporated using the vacuum pump to arrive at a neutral amphetamine base oil and combined with that from step 6.
  9. The neutral amphetamine base oil is dissolved in 2 L of dry acetone.
  10. Sulphuric acid (or phosphoric acid) is added drop by drop to reach a pH of 6 (in constant mixture).
  11. The mixture is then placed in a refrigerator for 12 hours.
  12. After the crystallisation procedure in the refrigerator, the amphetamine salt suspension is filtered through a Nutsche filter and then cleaned with cold dry acetone to obtain a whitish colour. The acetone from the cleaning procedure can be crystallised as described in steps 11-12 (but the quality of the amphetamine salt will be reduced). The yield rate is 60-70%.
